LB Agar Plates with Gentamicin-7, Kanamycin-50, Tetracycline-10, Bluo-gal-100, IPTG-40 are nutrient agar plates formulated for the growth and selection of antibiotic-resistant E. coli strains. The LB base supports routine cultivation, and the combined antibiotics enable selection of transformants carrying compatible gentamicin-, kanamycin-, or tetracycline-resistance markers.

Bluo-Gal and IPTG facilitate blue/white screening via lacZ α-complementation in suitable host strains, allowing visual discrimination of colonies with functional versus disrupted β-galactosidase activity when using appropriate cloning vectors in E. coli.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Kanamycin Sulfate
  • 7µg/mL Gentamicin Sulfate
  • 10µg/mL Tetracycline Hydrochloride
  • 100µg/mL Bluo-Gal
  • 40µg/mL Isopropyl-B-D-thiogalactopyranoside(IPTG)
  • Triple-Antibiotic Selection: Simultaneous selection of transformants carrying kanamycin-, gentamicin-, and tetracycline-resistance plasmids in E. coli.

  • Co-Transformation/Co-Maintenance: Stable maintenance of multiple compatible plasmids with distinct resistance markers in E. coli.

  • Blue/White Screening: lacZ α-complementation screening using Bluo-Gal and IPTG to distinguish recombinant (white) from non-recombinant (blue) colonies in E. coli.

  • Reporter-Based Library Screening: Visual screening of clone libraries where lacZ readout (Bluo-Gal + IPTG) and triple selection are required in E. coli.

  • Expression Construct Screening: Plate-level IPTG induction to assess phenotype or reporter activation from lac-promoter plasmids under triple-antibiotic pressure in E. coli.

  • Bacterial Two-Hybrid/Interaction Assays: Selection of multi-plasmid systems with lacZ reporters for blue colony readout in E. coli.

  • Strain and Plasmid Verification: Replica plating and confirmation of resistance profiles and insert presence via blue/white outcomes in E. coli.

  • Colony Isolation and Picking: Single-colony isolation, CFU enumeration, and downstream plasmid prep workflows under stringent selection in E. coli.

  • Stability Testing: Assessment of plasmid retention and compatibility under combined kanamycin, gentamicin, and tetracycline selection in E. coli.
