LB Agar Plates with Carbenicillin-100, X‑Gal, and IPTG are used for growth and selection of carbenicillin‑resistant E. coli strains carrying beta‑lactamase. Carbenicillin disodium salt at 100 µg/mL enables selection of plasmid‑bearing transformants.

The combination of 60 µg/mL X‑Gal and 0.1 mM IPTG supports blue‑white screening via lacZ α‑complementation: colonies expressing functional β‑galactosidase appear blue, whereas colonies with disrupted lacZα remain white.

Blue-White Screening: Differentiates recombinant from non-recombinant colonies via lacZ α-complementation on X-Gal/IPTG in E. coli lacZΔM15 strains (e.g., E. coli DH5α, JM109, TOP10, XL1-Blue).
Antibiotic Selection: Selects for bla/ampR plasmid-bearing E. coli using 100 µg/mL carbenicillin to reduce satellite growth versus ampicillin.
Cloning Workflows: Screens ligation, Gibson Assembly, and Golden Gate reactions by colony color and growth.
Library Screening: Identifies desired variants in cDNA, genomic, or mutagenesis libraries using colorimetric readout.
Colony Picking for Downstream Analysis: Isolates transformants for miniprep, restriction analysis, Sanger sequencing, or NGS.
Promoter/Reporter Assessment: Induces lac/T7-lac controlled reporters or constructs on solid media with 0.1 mM IPTG.
Plasmid Maintenance: Propagates ampicillin/carbenicillin-selectable plasmids in E. coli with reduced β-lactam degradation.
Transformation QC: Measures transformation efficiency of competent E. coli using blue-white and antibiotic selection readouts.
