LB Agar Plates with carbenicillin, X-Gal, and IPTG are a nutritionally balanced solid medium that supports the growth and selection of carbenicillin-resistant E. coli strains.

The combination of X-Gal and IPTG enables blue-white screening via lacZ α-complementation, allowing visual differentiation of recombinant and non-recombinant colonies on a single plate.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Carbenicillin Disodium Salt
  • 60µg/mL X-Gal
  • 0.1mM Isopropyl-B-D-thiogalactopyranoside(IPTG)
  • Blue-White Screening: Differentiates recombinant from non-recombinant colonies via lacZ α-complementation in E. coli using X-Gal and IPTG.

  • Antibiotic Selection: Selects and maintains plasmids bearing beta-lactam resistance in E. coli using carbenicillin.

  • Clone Library Screening: Screens ligation, Gibson Assembly, TA, or Golden Gate cloning libraries for inserts in E. coli.

  • Colony Picking and Verification: Enables efficient picking of white colonies for downstream colony PCR, restriction analysis, or sequencing.

  • Transformation Efficiency Assessment: Measures competent cell performance by CFU counts and blue/white ratios in E. coli.

  • Reporter and Promoter Testing: Assesses lac-based and IPTG-inducible promoter activity on plates in E. coli.

  • Plasmid Maintenance and Stability: Propagates and preserves plasmids with ampicillin-class selection in E. coli.

  • Strain Validation for α-Complementation: Verifies compatibility of host strains (e.g., lacZΔM15) and vectors carrying lacZα in E. coli.
