LB Agar Plates with 1% Glucose and Kanamycin-30 are a nutrient-rich agar medium that supports the growth and selection of kanamycin-resistant E. coli strains. The addition of 1% glucose enhances metabolic activity and biomass yield, and 30 µg/mL kanamycin sulfate enables effective selection of plasmid-bearing transformants.

  • 1% Tryptone
  • 0.5% Yeast extract
  • 1.0% Sodium chloride
  • 1.5% Agar
  • 1.0% Glucose
  • 30µg/mL Kanamycin Sulfate
  • Antibiotic Selection of Transformants: Isolation and maintenance of kanamycin-resistant colonies (30 µg/mL) in E. coli using LB (1% Tryptone, 0.5% Yeast extract, 1.0% NaCl, 1.5% Agar) with 1.0% Glucose.

  • Plasmid Maintenance: Stable propagation of KanR plasmids and expression constructs in E. coli and related Enterobacteriaceae.

  • Colony Isolation and CFU Enumeration: Streaking, picking, and counting kanamycin-resistant colonies for titering and purity checks.

  • Cloning/Subcloning Workflows: Recovery and screening of recombinant clones by colony PCR or restriction analysis from KanR colonies.

  • Basal Expression Suppression: 1.0% Glucose reduces catabolite-activated promoter leakiness (e.g., lac/ara systems) during clone recovery.

  • Genomic Integration Selection: Selection of strains carrying kanamycin-marked insertions, deletions, or transposons in E. coli.

  • Counter-Selection Against Contaminants: Inhibits growth of non-KanR microbes while supporting KanR strains.

  • Library Maintenance: Preservation and passaging of KanR plasmid libraries or mutant collections.

  • Conjugation/Strain Mating Selection: Selection of KanR transconjugants in systems using E. coli donors/recipients.
