LB Agar Plates with 1% Glucose and Ampicillin-100 are a nutrient agar medium that supports the growth and selection of ampicillin-resistant E. coli strains. The addition of 1% glucose enhances metabolic activity and biomass yield, and 100 µg/mL ampicillin sodium salt enables effective selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast extract
  • 1.0% Sodium chloride
  • 1.5% Agar
  • 1.0% Glucose
  • 100µg/mL Ampicillin Sodium Salt
  • Antibiotic Selection of Transformants: Selects for ampicillin-resistant plasmid-bearing Escherichia coli colonies using 100 µg/mL ampicillin.

  • Post-Transformation Plating: Recovers and isolates colonies after heat-shock or electroporation of E. coli with AmpR plasmids.

  • Single-Colony Isolation: Streaking to single colonies for clonal purification of AmpR strains.

  • Colony PCR Screening: Rapid verification of inserts or constructs directly from E. coli colonies.

  • Starter Culture Inoculation: Picking verified colonies to start liquid cultures for minipreps or expression.

  • CFU Enumeration with Selection: Quantifying viable AmpR cells via colony counting.

  • Plasmid and Strain Maintenance: Short-term storage and routine passaging of ampicillin-resistant E. coli clones.

  • Reduced Basal Expression: 1% glucose suppresses lac/T7-lac promoter leakiness during cloning of potentially toxic genes in E. coli.

  • Replica Plating: Confirming antibiotic resistance and phenotype consistency of transformed E. coli colonies.

  • Library Construction and Screening: Selecting members of plasmid libraries requiring ampicillin resistance in E. coli.
