LB Agar Plates with Kanamycin-50 and Streptomycin-50 are solid LB media formulated for growth and selection of kanamycin- and streptomycin-resistant E. coli. The plates contain 50 µg/mL kanamycin sulfate and 50 µg/mL streptomycin to provide dual antibiotic selection.

These plates are suitable for maintaining plasmids or strains carrying kanamycin and streptomycin resistance markers, enabling co-selection and routine propagation of engineered E. coli under dual selective conditions.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Kanamycin Sulfate
  • 50µg/mL Streptomycin
  • Plasmid Selection and Maintenance: Selects and maintains strains harboring kanamycin- and streptomycin-resistance plasmids in Escherichia coli using dual antibiotic pressure (50 µg/mL each).

  • Dual-Marker Co-Selection: Ensures retention of multi-plasmid systems or bicistronic constructs requiring both kanamycin and streptomycin resistance.

  • Clonal Isolation and Colony Purification: Streaks for single colonies on 1.5% agar LB to obtain pure, antibiotic-selected clones.

  • Transformation and Library Screening: Screens transformants or pooled libraries by colony growth under Kan50/Str50 selection.

  • Replica Plating and Stability Testing: Assesses plasmid stability and phenotype retention via replica plating on dual-antibiotic LB agar.

  • Conjugation/Mating Selection: Selects transconjugants after bacterial mating (e.g., Escherichia coli donor/recipient workflows) using dual resistance markers.

  • CRISPR and Genome Engineering Workflows: Maintains editing vectors and helper plasmids requiring kanamycin and streptomycin resistance.

  • Contamination Control in Cultures: Enriches for intended resistant strains while suppressing susceptible contaminants on solid LB medium.
