LB Agar Plates with Chloramphenicol-12.5 and Kanamycin-50 is a solid medium that supports growth and dual antibiotic selection of chloramphenicol- and kanamycin-resistant E. coli strains. The inclusion of 12.5 µg/mL chloramphenicol and 50.0 µg/mL kanamycin sulfate enables selection and maintenance of plasmid-bearing transformants with compatible resistance markers.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 12.5µg/mL Chloramphenicol
  • 50.0µg/mL Kanamycin Sulfate
  • Dual-Antibiotic Plasmid Selection: Selects transformants carrying chloramphenicol- and kanamycin-resistant plasmids in E. coli.

  • Co-Transformation Screening: Identifies colonies that have successfully taken up two compatible plasmids (CamR and KanR).

  • Plasmid Maintenance: Maintains dual-plasmid systems (e.g., expression vector + helper/regulatory plasmid) under stringent selection.

  • Library Construction and Cloning: Screens recombinant libraries where vectors require simultaneous chloramphenicol and kanamycin resistance.

  • Conjugation/Mobilization Assays: Selects recipients bearing both resistance markers after mating in E. coli or Agrobacterium tumefaciens.

  • Transposon/Genomic Integration Studies: Recovers strains with combined genomic and plasmid markers conferring CamR and KanR.

  • Protein Expression Workflows: Plates expression strains carrying a KanR expression plasmid and a CamR chaperone/regulatory plasmid.

  • Contamination Control: Reduces growth of unwanted microbes by requiring dual resistance while culturing engineered E. coli.

  • Synthetic Biology Constructs: Maintains multi-plasmid circuits with distinct selection markers for stable operation.

  • Strain Verification: Confirms retention of both antibiotic markers during strain banking and passaging.
