LB Agar Plates with Ampicillin-100 and Kanamycin-50 are nutrient agar media for growth and dual-antibiotic selection of ampicillin- and kanamycin-resistant E. coli strains.

Incorporated antibiotics at 100 µg/mL ampicillin sodium salt and 50 µg/mL kanamycin sulfate enable selection of plasmid-bearing transformants carrying beta-lactam and aminoglycoside resistance markers, including co-transformed cultures.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Ampicillin Sodium Salt
  • 50µg/mL Kanamycin Sulfate
  • Dual-Antibiotic Selection: Selects transformed E. coli carrying ampicillin- and kanamycin-resistance plasmids on LB agar (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl, 1.5% Agar; 100 µg/mL Ampicillin, 50 µg/mL Kanamycin).

  • Co-Transformation Maintenance: Maintains multiple compatible plasmids (e.g., helper and expression vectors) in E. coli under Amp/Kan dual selection.

  • Colony Isolation and Screening: Enables single-colony isolation for colony PCR, sequencing, and verification of recombinant E. coli with dual markers.

  • CRISPR/Recombineering Workflows: Supports stable propagation of CRISPR or recombineering plasmid systems requiring ampicillin and kanamycin selection in E. coli.

  • Starter Culture Preparation: Provides selective plates for picking verified colonies to inoculate liquid cultures for plasmid minipreps and expression.

  • Library Construction and Maintenance: Used for maintaining dual-marker plasmid libraries or combinatorial constructs in E. coli.

  • Strain Validation and QC: Confirms presence of Amp/Kan resistance markers to detect loss of plasmids or contamination in E. coli stocks.

  • Genetic Transfer Workflows: Selects desired recipients or transductants in E. coli after conjugation or transduction when Amp/Kan markers are employed.
