LB Agar Plates with 10 mM Arabinose and Ampicillin-50 is a nutrient agar medium used for cultivation and selection of ampicillin-resistant E. coli strains.

The inclusion of 10 mM L-arabinose enables induction of PBAD-regulated expression systems, and 50 µg/mL ampicillin sodium salt maintains selection for plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Ampicillin
  • 10mM Arabinose
  • Ampicillin-Resistant Transformant Selection: Plates Escherichia coli transformants carrying bla/ampR plasmids at 50 µg/mL ampicillin.

  • PBAD/araC-Induced Expression Screening: Enables robust plate-based induction of genes under the arabinose-inducible PBAD promoter (10 mM arabinose) in Escherichia coli.

  • Reporter Assays (e.g., GFP/RFP): Visual screening of PBAD-driven fluorescent or colorimetric reporters on colonies of Escherichia coli.

  • Plasmid Maintenance Under Selection: Sustains PBAD/araC plasmids with ampicillin selection during streaking, isolation, and storage of Escherichia coli strains.

  • Toxic Gene Expression on Plates: Induces controlled, colony-level expression of otherwise burdensome or toxic genes from PBAD in Escherichia coli.

  • Colony Screening and Picking: Rapid isolation of correctly transformed Escherichia coli colonies for downstream colony PCR, minipreps, and sequencing.

  • Recombineering/Genome Editing Workflows: Supports arabinose-inducible systems (e.g., PBAD-driven recombinases or editing components) during selection in Escherichia coli.

  • CRISPRi/CRISPRa Induction Tests: Plate-level induction and assessment of PBAD-controlled dCas9 or guide expression in Escherichia coli.
