LB Agar Plates with Tetracycline-5 are general-purpose agar plates formulated for the growth and selection of tetracycline-resistant E. coli strains. The inclusion of 5 µg/mL tetracycline hydrochloride enables selection and maintenance of tetracycline-selectable plasmids.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 5µg/mL Tetracycline Hydrochloride
  • Antibiotic-Selective Cloning: Selection and isolation of transformants carrying tetracycline-resistance plasmids in E. coli (5 µg/mL tetracycline for efficient colony recovery).

  • Plasmid Maintenance: Stable propagation of tet-marked plasmids and genomic constructs in E. coli and Bacillus subtilis under antibiotic pressure.

  • Transformation and Efficiency Plating: Quantifying transformation efficiency and recovery of competent cells on tetracycline-selective LB agar.

  • Recombinant Screening: Colony screening (e.g., colony PCR or blue-white screening with IPTG/X-gal) for inserts on vectors conferring tetracycline resistance.

  • Strain Preservation: Preparing clonal isolates and glycerol stocks from single colonies while maintaining tet-linked genetic elements.
