LB Agar Plates with Kanamycin-20 are solid nutrient media for cultivation and selection of kanamycin-resistant E. coli strains. The LB base provides peptides, amino acids, vitamins, and sodium chloride for routine growth, while 1.5% agar solidifies the medium for colony isolation. Incorporation of 20 µg/mL kanamycin sulfate enables selection of plasmid-bearing transformants carrying kanamycin resistance markers.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 20µg/mL Kanamycin Sulfate
  • Selective Cultivation of Kanamycin-Resistant Bacteria: Growth and maintenance of kanamycin-resistant strains (e.g., Escherichia coli, Agrobacterium tumefaciens, Bacillus subtilis) carrying kan-marked plasmids.

  • Transformation Screening: Selection of recombinant colonies after heat-shock or electroporation when vectors encode kanamycin resistance.

  • Colony Isolation and Picking: Streaking for single colonies and picking stable clones for downstream plasmid prep and sequencing.

  • Plasmid Maintenance: Routine passaging to retain kan-marked plasmids and minimize plasmid loss in Escherichia coli.

  • CFU Enumeration Under Selection: Titering and colony counting of kan-resistant populations to assess transformation efficiency or viability.

  • Conjugation and Transposon Selections: Post-mating or mutagenesis selection where kanamycin resistance serves as the selectable marker.

  • Replica Plating for Resistance Verification: Confirming plasmid carriage and antibiotic susceptibility profiles across colonies.
