LB Agar Plates with Apramycin-50 is a solid medium used for cultivation and selection of apramycin-resistant E. coli and other Gram-negative bacteria. The formulation provides general nutrients and solid support suitable for routine cloning workflows.

Apramycin at 50 µg/mL enables selection of plasmid-bearing transformants and maintenance of apramycin-resistance markers during colony growth and screening.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Apramycin
  • Apramycin-Resistant Clone Selection: Selects and maintains plasmids or genomic integrations carrying aac(3)IV in Escherichia coli and other Gram-negative bacteria at 50 µg/mL.

  • Transformation Screening: Plates and screens recombinant transformants in E. coli using apramycin selection on LB (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl, 1.5% Agar).

  • Intergeneric Conjugation Selection: Recovers apramycin-resistant exconjugants in Streptomyces following mating with E. coli donors.

  • Library Maintenance: Maintains BAC/cosmid/fosmid libraries with apramycin resistance markers in E. coli.

  • Genome Editing Selection: Selects CRISPR/Cas or recombineering edits that use apramycin resistance cassettes.

  • Allelic Exchange and Transduction: Selects marker exchange events or P1 transductants carrying apramycin resistance in E. coli.

  • Transposon Mutagenesis: Recovers mutants from Tn-based insertions with apramycin markers in E. coli and Pseudomonas.

  • Strain Authentication and Stability: Verifies plasmid retention and antibiotic resistance stability by streaking and colony isolation.

  • CFU Enumeration Under Selection: Quantifies viable counts of apramycin-resistant cells from mixed cultures.

  • Pathogen and Commensal Workflows: Selects engineered strains of Salmonella, Klebsiella, and other Enterobacteriaceae carrying apramycin markers.
