LB Agar Plates with Gentamicin-30 are nutrient agar plates for cultivation and selection of gentamicin-resistant E. coli and related host strains. The medium provides general nutritional support for routine cloning and strain propagation, and 30 µg/mL gentamicin sulfate enables selection of plasmid-bearing transformants carrying a gentamicin resistance marker.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 30µg/mL Gentamicin Sulfate
  • Antibiotic Selection of Transformants: Isolates colonies carrying gentamicin-resistance plasmids (30 µg/mL) in Escherichia coli and other Gram-negative bacteria.

  • Plasmid Cloning and Maintenance: Maintains vectors with gentamicin-resistance cassettes (e.g., aacC1/aac(3)) in Escherichia coli.

  • Colony Isolation and Streaking: Obtains single colonies under gentamicin selection for downstream workflows.

  • CFU Enumeration Under Selection: Quantifies viable gentamicin-resistant cells after transformation or conjugation.

  • Transposon Mutant Selection: Recovers mutants carrying gentamicin-marked transposons in organisms such as Escherichia coli and Pseudomonas aeruginosa.

  • Bacterial Conjugation/Mating Selection: Selects recipients or exconjugants acquiring gentamicin-marked plasmids.

  • Library Maintenance: Preserves genomic or plasmid libraries that use gentamicin resistance as the selectable marker.

  • Plasmid Stability Screening: Assesses retention of gentamicin-marked plasmids during serial passaging.

  • Phenotypic Verification of Resistance: Confirms expected gentamicin-resistance phenotypes via spot or streak assays.
