LB Agar Plates with Carbenicillin-200 is an agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The inclusion of 200 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants on a standard LB nutrient base.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 200µg/mL Carbenicillin Disodium Salt
  • Bacterial Transformation and Selection: Selects transformants carrying bla-marked plasmids on LB (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl, 1.5% Agar) with 200 µg/mL carbenicillin.

  • Plasmid Maintenance and Propagation: Routine streaking and passaging of Escherichia coli strains harboring beta-lactamase (bla) plasmids.

  • Colony Screening and Clone Isolation: Picking single colonies for miniprep, restriction analysis, PCR, or sequencing of bla-bearing constructs.

  • Starter Culture Preparation: Inoculating isolated antibiotic-selected colonies for overnight cultures used in cloning or protein expression.

  • Transformation Efficiency and CFU Enumeration: Quantifying colony-forming units under 200 µg/mL carbenicillin selection.

  • CRISPR and Genome Editing Workflows: Selecting edited clones that retain carbenicillin-resistant editing or helper plasmids.

  • Conjugation/Mobilization Experiments: Selecting recipients that acquired bla-marked plasmids after mating.

  • Library Maintenance: Preserving pooled plasmid libraries in E. coli under beta-lactam selection to minimize plasmid loss.

  • Strain Archiving and Recovery: Streaking glycerol stocks of E. coli carrying bla vectors to verify viability and plasmid retention.

  • Antibiotic Susceptibility Controls: Verifying beta-lactamase-mediated resistance versus sensitive parental strains on a defined LB-carb background.
