LB Lennox Agar Plates with Zeocin-50 are solid LB Lennox medium formulated for cultivation and antibiotic selection of Zeocin®-resistant bacteria. The medium provides a consistent nutrient base for routine cloning and maintenance of recombinant strains, including E. coli.

Zeocin® at 50 µg/mL enables selection of transformants carrying vectors with a ble resistance cassette, facilitating maintenance of plasmids under controlled selective pressure.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Zeocin® (an InvivoGen's product)
  • Selection of Zeocin-resistant transformants: Plating heat-shock or electroporated Escherichia coli on selective media to recover clones carrying Zeocin®-resistance plasmids.

  • Maintenance of Zeocin-selectable plasmids: Streaking and passaging Zeocin-marked constructs in E. coli and other susceptible bacteria to preserve genotype.

  • Clonal isolation for downstream analysis: Picking single Zeocin-resistant colonies for colony PCR, sequencing, and plasmid minipreps.

  • Transformation efficiency and CFU quantification: Counting colony-forming units under Zeocin selection to assess cloning or transformation success.

  • Library construction and screening: Selecting genetic libraries (e.g., mutagenesis or assembly pools) under Zeocin to enrich desired constructs.

  • Counter-selection of non-recombinants: Eliminating background cells lacking the Zeocin-resistance marker in mixed or ligation reactions.

  • Conjugation/transconjugant selection: Selecting Zeocin-marked transconjugants after bacterial mating (e.g., Escherichia coli, Agrobacterium tumefaciens).

  • CRISPR/recombineering workflows: Maintaining helper or editing plasmids with Zeocin resistance during genome engineering in E. coli.
