LB Agar Plates with Tetracycline-15 are a lysogeny broth agar medium formulated for cultivation and selection of tetracycline-resistant E. coli strains.

Tetracycline is present at 15 µg/mL to enable selection and maintenance of plasmids bearing tetracycline resistance markers in E. coli cultures.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 15µg/mL Tetracycline

  • Antibiotic Selection of Transformants: Isolates colonies harboring tetracycline-resistant plasmids in Escherichia coli and other TetR hosts.

  • Plasmid Maintenance and Propagation: Sustains selective pressure to retain Tet-marked plasmids during routine culture and passaging.

  • Colony Isolation and Purification: Enables streaking and single-colony picking to obtain clonal Tet-resistant strains.

  • CFU Enumeration Under Selection: Quantifies viable Tet-resistant cells after transformation, conjugation, or recovery steps.

  • Clone Screening (e.g., Colony PCR): Supports screening of recombinant constructs from Tet-resistant colonies.

  • Conjugation/Mating Selection: Selects transconjugants carrying Tet-marked plasmids or transposons in donor/recipient systems (e.g., Escherichia coli, Bacillus subtilis).

  • Short-Term Strain Preservation: Maintains Tet-resistant strains on plates for working stocks.

  • Phenotypic Characterization on Selective Media: Assesses colony morphology and growth traits of Tet-resistant variants.
