LB Agar Plates with Kanamycin-30 are agar medium for cultivation and selection of kanamycin-resistant strains of gram-negative bacteria such as E. coli. The inclusion of 30 µg/mL kanamycin sulfate provides selective conditions for maintenance of kanamycin-resistance markers and recovery of plasmid-bearing transformants.

These plates are suitable for routine streaking, colony isolation, and cloning workflows that require selection with kanamycin.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 30µg/mL Kanamycin Sulfate
  • Antibiotic Selection of Transformants: Isolates kanamycin-resistant colonies after transformation of E. coli or other bacteria with kanR plasmids (30 µg/mL Kanamycin Sulfate).

  • Plasmid Maintenance and Propagation: Maintains selection for kanamycin-marked plasmids during routine streaking and passaging of E. coli.

  • Colony Screening for Cloning: Enables picking colonies for PCR, restriction analysis, and sequencing of kanR constructs.

  • Protein Expression Workflows: Selects kanR expression vectors in strains like E. coli BL21(DE3) prior to induction.

  • Conjugation and Mating Assays: Selects kanamycin-marked transconjugants in systems such as Agrobacterium tumefaciensE. coli or Pseudomonas aeruginosa.

  • CRISPR/Gene Knockout Selection: Screens for insertions or replacements using kanamycin resistance cassettes in E. coli and other bacteria.

  • Transposon Mutagenesis: Selects kanR transposon insertions in organisms including E. coli and Bacillus subtilis.

  • Colony-Forming Unit (CFU) Enumeration: Quantifies viable kanamycin-resistant bacteria under defined selection pressure.

  • Strain Verification and Stability Checks: Confirms retention of kanamycin markers by re-streaking and phenotype validation.
