LB Agar Plates with Chloramphenicol-25 is a nutrient-rich agar medium that supports the growth and selection of chloramphenicol-resistant E. coli strains and other bacteria commonly maintained on LB. The inclusion of 25 µg/mL chloramphenicol enables selection of plasmid-bearing transformants carrying chloramphenicol resistance markers.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 25µg/mL Chloramphenicol
  • Antibiotic Selection of Transformants: Isolates colonies carrying chloramphenicol-resistance plasmids (25 µg/mL) after transformation of Escherichia coli or Bacillus subtilis.

  • Cloning Workflows: Screens colonies after ligation, Gibson, or Golden Gate assembly by growth on selective LB agar.

  • Colony Isolation and Purification: Streaks for single colonies and purifies strains on chloramphenicol-selective plates.

  • Colony PCR Screening: Picks chloramphenicol-resistant colonies for insert verification by colony PCR.

  • Plasmid Maintenance: Maintains low- and medium-copy chloramphenicol-resistant plasmids (e.g., p15A, pACYC) in Escherichia coli under selection.

  • Strain Archiving and Recovery: Revives glycerol stocks and preserves engineered strains on selective plates.

  • CFU Enumeration on Selective Media: Quantifies viable chloramphenicol-resistant cells following transformations or selections.

  • Supplemented Screening: Supports add-on overlays (e.g., X-gal/IPTG) for phenotype screening of CmR constructs in Escherichia coli.
