LB Agar Plates with Chloramphenicol-15 is a Luria-Bertani agar medium that supports the growth and selection of chloramphenicol-resistant E. coli strains. The medium is solidified with agar and supplemented with chloramphenicol at 15 µg/mL to enable selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 15µg/mL Chloramphenicol
  • Selective Plating of Transformants: Recovery of colonies carrying chloramphenicol-resistant plasmids (e.g., cat-marked vectors, BACs, cosmids) in Escherichia coli.

  • Plasmid Maintenance: Routine culture and passaging to retain chloramphenicol-selectable plasmids in Escherichia coli expression and cloning strains.

  • Cloning and Library Screening: Colony isolation and picking during molecular cloning and library construction under chloramphenicol selection.

  • Streaking for Single Colonies: Strain purification and isolation of single colonies while maintaining chloramphenicol selection.

  • CFU Enumeration: Determination of viable cell counts for chloramphenicol-resistant cultures.

  • Replica Plating: Verification of chloramphenicol resistance or plasmid presence across multiple plates.

  • Conjugation/Mating Selections: Post-mating selection of chloramphenicol-resistant transconjugants.

  • Genome Editing Selections: Screening of clones in CRISPR/recombineering workflows when cat is used as the selectable marker.

  • Protein Expression Workflows: Maintenance of strains harboring chloramphenicol-selectable expression vectors prior to induction.

  • Quality Control and Strain Verification: Selective plating to confirm genotype, antibiotic resistance, and detect contamination.
