LB (Luria–Bertani) Agar with Carbenicillin-150 is a solid nutrient medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The inclusion of 150 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants.

  • 1.00% Tryptone
  • 0.50% Yeast Extract
  • 1.00% Sodium Chloride
  • 1.50% Agar
  • 150µg/mL Carbenicillin Disodium Salt
  • Antibiotic Selection of Plasmid-Containing Escherichia coli: Selects for β-lactamase–encoded resistance using 150 µg/mL carbenicillin for robust plasmid maintenance.

  • Molecular Cloning Workflows: Plating transformants after ligation or Gibson assembly to recover recombinant clones.

  • Blue-White Screening (with X-gal/IPTG): Distinguishes insert-containing colonies on LB-carbenicillin when α-complementation vectors are used.

  • Colony PCR and Sanger Prep: Picking colonies for rapid insert verification directly from selective plates.

  • Transformation Efficiency and CFU Enumeration: Quantifying transformants and optimizing competent cell performance under carbenicillin selection.

  • Protein Expression Strain Selection: Maintaining expression plasmids in T7-based or other E. coli systems prior to induction.

  • CRISPR/Cas Genome Editing Workflows: Selecting cells carrying editing or helper plasmids conferring carbenicillin resistance.

  • Bacterial Conjugation/Plasmid Transfer Selection: Selecting transconjugants when the mobilized plasmid carries a carbenicillin resistance marker.

  • Glycerol Stock Preparation: Isolating single colonies for long-term storage of carbenicillin-resistant E. coli strains.

  • Routine Plasmid Maintenance: Preserving low- to high-copy vectors in laboratory E. coli strains under consistent β-lactam selection.
