LB Agar Plates with Carbenicillin-75 are solid nutrient media that support the growth and selection of carbenicillin-resistant E. coli strains. The medium supplies peptides, amino acids, vitamins, and sodium chloride for cultivation, and 75 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 75µg/ml Carbenicillin Disodium Salt
  • Selection of Transformants: Isolates carbenicillin-resistant Escherichia coli colonies after heat-shock or electroporation transformation of β-lactamase–encoded plasmids (75 µg/mL carbenicillin).

  • Clonal Isolation and Maintenance: Streaks for single colonies and maintains plasmid-bearing E. coli strains on LB agar (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl, 1.5% Agar).

  • Colony Screening: Enables colony PCR, restriction analysis, or sequencing from selected E. coli clones carrying ampicillin/carbenicillin-resistance plasmids.

  • Blue-White Screening (with supplements): Supports α-complementation screening of E. coli when supplemented with X-gal/IPTG.

  • CFU Enumeration: Quantifies transformation efficiency by plating serial dilutions and counting colonies under carbenicillin selection.

  • Plasmid Stability Assays: Assesses segregational stability of β-lactamase–marked plasmids in E. coli under 75 µg/mL carbenicillin.

  • Library Plating: Plates combinatorial or genomic libraries in E. coli to ensure selectable coverage of transformants.

  • Replica Plating: Verifies antibiotic resistance markers and clone identity by transferring colonies onto selective media.

  • Strain Verification: Confirms plasmid presence/expression in engineered E. coli strains prior to liquid culture scale-up.

  • Glycerol Stock Recovery: Recovers and re-isolates clonal E. coli lines from frozen stocks under carbenicillin selection.
