LB Agar Plates with Carbenicillin-50 is a solid agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The LB nutrient base provides peptides, amino acids, vitamins, and ionic balance for general cultivation, and 50 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Carbenicillin Disodium Salt
  • Plasmid Selection and Maintenance: Selects and maintains bla (β-lactamase)-bearing plasmids in Escherichia coli using 50 µg/mL carbenicillin.

  • Bacterial Transformation Recovery and Screening: Enables post-transformation growth and selection of recombinant E. coli colonies carrying carbenicillin-resistant plasmids.

  • Cloning Workflows (Ligation/Gibson/Golden Gate): Supports selection of correctly assembled constructs on solid LB medium.

  • Colony Isolation and Purification: Streaking for single colonies to derive clonal, antibiotic-selected strains.

  • Colony PCR and Mini-Prep Pre-Screening: Picking antibiotic-resistant colonies for rapid insert verification.

  • Blue-White Screening (with X-gal/IPTG): Used for alpha-complementation screening when supplemented with chromogenic inducers.

  • CFU Enumeration Under Selection: Quantifying transformation efficiency and viable counts of resistant E. coli on selective plates.

  • Pre-Expression Strain Qualification: Verifying plasmid carriage in protein expression strains prior to induction.

  • Routine Strain Storage and Propagation: Maintaining glycerol stock derivatives and working plates with selective pressure.

  • Reduced Satellites vs. Ampicillin: Preferentially used to minimize satellite colony formation during selection on LB agar.
