LB Agar with Puromycin-100 is a nutrient agar medium that supports the growth and selection of puromycin-resistant bacterial populations. The matrix provides peptides, amino acids, vitamins, and appropriate ionic strength for routine culture, and agar supplies a firm surface for colony isolation.

Puromycin dihydrochloride at 100 µg/mL enables selection of cells expressing puromycin N-acetyltransferase (pac) by inhibiting protein synthesis in non-resistant cells.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Puromycin Dihydrochloride
  • Selection of Puromycin-Resistant Clones: Isolating bacterial transformants carrying pac-encoded puromycin resistance on LB agar with 100 µg/mL puromycin.

  • Plasmid Maintenance in E. coli Hosts: Propagation of pac-containing plasmids in strains such as E. coli DH5α, TOP10, Stbl3, and BL21(DE3).

  • Colony Screening and Picking: Selecting recombinant colonies for cloning workflows under puromycin pressure.
