20X HBS-EP+, pH 7.6 is a HBS-EP+ concentrate in purified water. Dilution to 1X gives 10 mM HEPES, 150 mM sodium chloride, 3 mM EDTA, and 0.05% polysorbate 20 (Tween-20) at pH 7.6. The 1 L fill makes 20 L of 1X buffer.

It is commonly used to prepare running buffer for SPR binding and kinetics studies of proteins, antibodies, and other biomolecules. The higher surfactant level than HBS-EP further reduces nonspecific binding. Dilute 50 mL to 1 L with purified water and degas before use.

  • 200mM HEPES Free Acid
  • 60mM EDTA
  • 1.0% Tween-20
  • SPR Running Buffer: HEPES-buffered saline with EDTA and surfactant for SPR binding and kinetics studies of antibodies and ADCs.
  • ELISA Plate Washing: HEPES-buffered saline with surfactant for washing plates between assay steps.
  • Biophysical and Stability Studies: HEPES-buffered saline with surfactant for binding and stability measurements of proteins.
  • SEC Mobile Phase: Mobile phase for size-exclusion chromatography of antibody and ADC aggregates and fragments.
  • Enzyme Assays: HEPES-buffered saline for enzyme and binding assays near neutral pH.
  • Sample and Reagent Dilution: Diluent for samples, standards, and reagents.
  • Sample Handling and Storage: HEPES-buffered saline for holding and diluting protein samples.
  • Protein Purification and Chromatography: HEPES-buffered saline for purification and holding steps near neutral pH.
