Glycine, pH 2.7, is an acidic buffering solution used to elute antibodies from Protein A/G affinity resins and to dissociate antigen–antibody complexes in immunochemical workflows.

It is applied for eluting bound immunoglobulins, stripping antibodies from cell or microbial surfaces (e.g., E. coli), and regenerating affinity matrices following capture steps.

Following elution, immediate neutralization is typically performed to limit acid exposure of proteins. The buffer is suitable for downstream processes such as buffer exchange, SDS-PAGE, and immunoblotting.

  • 0.1M Glycine
  • Antibody Elution (Protein A/G/L Chromatography): Elutes bound IgG with low-pH conditions using 0.1M Glycine, pH 2.7, followed by rapid neutralization.

  • Immunoprecipitation (IP) Elution: Releases antigen–antibody complexes from beads with 0.1M Glycine, pH 2.7, preserving target integrity.

  • Western Blot Antibody Stripping: Removes primary/secondary antibodies from membranes using acidic 0.1M Glycine for reprobing.

  • Formaldehyde Crosslink Quenching (ChIP/Crosslinking): Stops fixation reactions by quenching reactive aldehydes with 0.1M Glycine.

  • NHS-Ester Labeling Quench: Terminates amine-reactive (NHS) conjugation reactions and reduces background using 0.1M Glycine.

  • Ligand–Receptor Dissociation Assays: Employs transient low pH (0.1M Glycine, pH 2.7) to dissociate surface-bound complexes for recycling studies.

  • Phage Display/Biopanning Elution: Uses low-pH 0.1M Glycine to elute bound phage from targets during selection cycles.

  • Protein Purification Cleanup: Removes nonspecific proteins from affinity matrices via brief acidic washes with 0.1M Glycine.

  • Sample Neutralization/Conditioning Steps: Provides controlled pH shifts before immediate neutralization in workflows requiring transient acidification.

  • Membrane and Bead Regeneration: Applies low pH (0.1M Glycine, pH 2.7) to regenerate affinity media between runs.
