1 M Glycine Buffer, pH 3.2, is an acidic, zwitterionic buffer suitable for procedures requiring low pH conditions. The solution maintains pH near the carboxyl pKa of glycine and provides high ionic strength for controlled pH adjustment in biochemical workflows.

Applications include elution of antibodies from Protein A/G affinity matrices, dissociation of immune complexes, stripping of proteins from solid supports, pH-shift elution in chromatography, sample acidification followed by neutralization, and pH control in electrophoresis and enzyme inactivation steps.

  • Protein A/G Affinity Chromatography Elution: Acidic elution of antibodies and Fc-fusion proteins; commonly diluted from 1M to 0.1M glycine, pH ~2.7–3.2, then neutralized.

  • Western Blot/Membrane Reprobing: Elution of bound primary/secondary antibodies from membranes to enable reprobing.

  • Immune Complex Dissociation: Acidic dissociation of antigen–antibody complexes prior to analysis (e.g., ELISA, immunoprecipitation).

  • Cell Surface Antibody Stripping: Rapid removal of surface-bound antibodies from live cells for receptor internalization/recycling assays.
