CM Glucose Plates minus Histidine and Leucine are defined agar plates for the maintenance and selection of yeast strains using histidine and leucine auxotrophic markers. Glucose serves as the primary carbon source and ammonium sulfate supplies inorganic nitrogen.

The omission of histidine and leucine restricts growth to strains that synthesize these amino acids or carry complementing markers (e.g., HIS3 and LEU2) on plasmids or integrated constructs, enabling selection and maintenance of targeted genotypes in Saccharomyces cerevisiae and related yeasts.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • Selection of S. cerevisiae transformants (LEU2, HIS3): Isolates yeast carrying LEU2 and HIS3 auxotrophic markers on defined 2.0% glucose, 0.5% ammonium sulfate plates.

  • Yeast two-hybrid workflows: Maintains bait/prey strains with LEU2 and HIS3 markers and supports interaction-dependent growth on histidine dropout.

  • Plasmid maintenance and stability assays: Ensures retention of LEU2/HIS3-marked plasmids under dual auxotrophic selection.

  • CRISPR/Cas9 genome editing selection: Screens edits using LEU2 and HIS3 selectable markers in glucose-defined minimal conditions.

  • Replica plating and spot assays: Compares growth phenotypes and mutant fitness on defined glucose medium lacking histidine and leucine.

  • Genetic crosses and tetrad analysis: Scores segregation of LEU2/HIS3 genotypes on selective dropout plates.

  • Library and suppressor screens: Selects interaction/suppressor hits requiring LEU2 and HIS3 auxotrophy complementation.

  • Metabolic and signaling studies under glucose repression: Assesses pathways in fermentative conditions with defined nitrogen (ammonium sulfate) while selecting for LEU2/HIS3.

  • Strain authentication and backcrossing: Confirms marker presence and linkage using dual dropout selection in defined medium.
