1 M Calcium Chloride is a concentrated aqueous salt solution that supplies calcium ions for molecular biology and microbiology workflows. It is commonly used for preparing chemically competent E. coli for heat-shock transformation, supplementing buffers that require divalent cations, stabilizing cell envelopes, and supporting protocols that employ calcium-dependent nucleases or membrane-associated processes.

This solution is suitable for dilution to working concentrations in transformation, phage handling, and precipitation workflows, and can be sterile-filtered and stored at 2–8 °C to maintain solution integrity. Thawing and mixing before use ensures homogeneity of the calcium ion concentration.

  • 1M Calcium Chloride Dihydrate

  • Chemically Competent Cell Preparation: Used to prepare CaCl2-competent E. coli by promoting DNA binding to the cell surface.

  • Heat-Shock Transformation: Facilitates plasmid uptake during CaCl2 heat-shock transformation of E. coli.

  • Bacteriophage Adsorption: Enhances adsorption and infection efficiency of phages (e.g., T4) in E. coli cultures.

  • Calcium Phosphate Transfection: Provides Ca2+ to form DNA–calcium phosphate precipitates for mammalian cell transfection.

  • Alginate Gelation: Crosslinks sodium alginate to form calcium alginate beads and hydrogels for encapsulation.

  • Cell Culture Supplementation: Adjusts extracellular Ca2+ levels in mammalian media to support adhesion and signaling.

  • Enzyme Assays: Supplies Ca2+ cofactor for calcium-dependent enzymes (e.g., DNase I, proteases, amylases).

  • Protein Crystallization: Used as a salt component in crystallization screens to promote crystal formation.
