Reinforced Clostridial Agar Plates (pH 6.8) are a nutritionally enriched solid medium for cultivation, maintenance, and enumeration of anaerobic clostridia, including Clostridium spp. The formulation combines complex nitrogen sources, yeast components, sodium chloride, a fermentable carbohydrate, acetate, starch, and a reducing agent to support recovery of stressed cells and growth under anaerobic conditions. The medium is solidified with agar.

Typical applications include culture of Clostridium perfringens, Clostridium sporogenes, and related species from environmental, food, and clinical specimens when used with appropriate anaerobic techniques. Plates are adjusted to pH 6.8 at 25 °C and are intended for general microbiological use.

  • 0.50% Tryptone
  • 1.00% Beef Extract
  • 0.50% Sodium Chloride
  • 0.30% Yeast Extract
  • 0.50% Proteose Peptone #3
  • 0.50% Glucose
  • 0.50% Sodium Acetate
  • 0.10% Starch
  • 0.06% Cysteine-HCl
  • 2.10% Agar
  • Anaerobic Isolation and Cultivation: Supports growth of Clostridium spp. from clinical, food, and environmental samples using a reducing formulation (0.06% Cysteine-HCl) and rich nutrients (peptones, extracts, 0.50% Glucose).

  • Enumeration of Anaerobes: Used to count clostridia and mixed anaerobic flora on plates with consistent pH 6.8 and 2.10% Agar for colony isolation.

  • Spore Recovery After Stress: Facilitates germination and recovery of heat- or chemically-stressed clostridial spores with 0.50% Sodium Acetate and 0.10% Starch supporting repair and growth.

  • Food and Dairy Testing: Employed for detection and enumeration of sulfite-reducing clostridia, including Clostridium perfringens, in process and finished products.

  • Pre-Enrichment Prior to Selective Media: Non-selective recovery medium before plating to selective/differential media for Clostridium spp., including Clostridioides difficile.

  • Maintenance and Subculture of Anaerobic Stocks: Used to maintain viability and purity of reference and working cultures of Clostridium spp.

  • Environmental Monitoring for Anaerobic Contaminants: Plates used in monitoring programs to detect clostridial contamination on surfaces, air (settle plates), and water concentrates.

  • Fermentation and Metabolic Studies: Suitable for assessing colony morphology and basic fermentation traits in clostridia (e.g., Clostridium acetobutylicum).
