Brain Heart Infusion Agar with Kanamycin-25 is a nutrient-rich agar medium that supports the cultivation of a broad range of bacteria and the selection of kanamycin-resistant strains. It is suitable for maintaining organisms such as E. coli carrying kanamycin resistance markers.

The inclusion of 25 µg/mL kanamycin sulfate provides antibiotic selection pressure for plasmid-bearing transformants, while the Brain Heart Infusion base supports growth of both nonfastidious and select fastidious bacteria.

  • 3.7% Brain Heart Infusion
  • 1.5% Agar
  • 25µg/mL Kanamycin Sulfate

  • Selection of Kanamycin-Resistant Transformants: Isolates colonies carrying kanamycin-resistance plasmids after transformation of Escherichia coli.

  • Plasmid Maintenance and Cloning: Maintains selection pressure for kanR plasmids during cloning and strain propagation in E. coli.

  • Transposon Mutagenesis Selection: Selects mutants harboring kan-marked transposons in E. coli, Salmonella enterica, or Pseudomonas aeruginosa.

  • Bacterial Conjugation/Mating Selections: Recovers transconjugants carrying kanamycin resistance markers.

  • CFU Enumeration of Kan-Resistant Strains: Quantifies viable counts of engineered or marked bacteria under kanamycin selection.

  • Isolation of Kan-Resistant Gram-Positive Strains: Supports growth and selection of kanR derivatives of Staphylococcus aureus, Bacillus subtilis, or Enterococcus faecalis.

  • Screening and Colony Picking: Enables reliable recovery of kanR colonies for downstream PCR, sequencing, and minipreps.

  • Genetic Recombineering/Editing Selections: Selects clones bearing kan-marked cassettes after recombineering or CRISPR-mediated edits.

  • Phage and Mobile Element Workflows: Maintains selection for kan-marked lysogens or integrants during phage or ICE experiments.

  • Quality Control of Kanamycin Selection: Serves as a positive-control medium to verify 25 µg/mL kanamycin activity against non-resistant strains.
