Cesium chloride is a high-density inorganic salt used to prepare isopycnic density gradients for nucleic acid and viral particle separations in ultracentrifugation. It is applied to fractionate DNA by buoyant density, distinguish closed-circular, nicked, and linear DNA in the presence of ethidium bromide, and to purify bacteriophages and viruses based on intrinsic density differences.

Typical workflows include plasmid DNA preparations from E. coli, separation of genomic DNA from plasmid DNA and RNA, and recovery of phage particles following amplification. The material is water-soluble and suitable for preparing gradients across a range of densities required for nucleic acid and viral purification protocols.

  • Plasmid DNA Purification: Uses Cesium Chloride (1.4 g/mL) with ethidium bromide for isopycnic ultracentrifugation to isolate supercoiled plasmids from Escherichia coli.

  • Viral Vector Purification: Creates equilibrium density gradients with Cesium Chloride (1.4 g/mL) to purify AAV and adenovirus particles from protein and nucleic acid contaminants.
