1X PBS-MK is a phosphate-buffered saline with added magnesium and potassium ions used for routine preparation, washing, dilution, and storage of cells and reagents in microbiology and molecular biology workflows. The buffer provides physiological ionic strength and pH suitable for general laboratory use.

Common applications include handling of E. coli cultures, maintenance of cell samples during processing, and preparation of solutions for protocols that require defined monovalent and divalent cations.

  • 1X PBS
  • 1mM Magnesium Chloride
  • 2.5mM Potassium Chloride

  • Cell Washing and Reagent Dilution: Isotonic 1X PBS with 1 mM MgCl2 and 2.5 mM KCl for maintaining cell integrity during handling.

  • Flow Cytometry/Cell Sorting: Supports cell viability and surface interactions; reduces clumping during acquisition.

  • Immunostaining (IHC/ICC) Washes: Gentle washes that preserve morphology and epitope integrity during antibody incubations.

  • DNase I Treatment: Provides Mg2+ cofactor for efficient genomic DNA removal in RNA preps and lysates.

  • Viral/Bacteriophage Assays: Mg2+ enhances phage adsorption and plaque formation on bacterial lawns.

  • Microbial Suspension/Dilution: Isotonic buffer for resuspending colonies and preparing inocula without lysing cells.

  • Live-Cell Microscopy: Stable pH/osmolarity buffer for short-term imaging of adherent and suspension cells.

  • Bead-Based and Receptor-Ligand Assays: Maintains divalent-cation–dependent interactions during binding and wash steps.
