15% Iodixanol is an iso-osmotic density medium intended for the preparation of continuous or discontinuous gradients for the fractionation of cells, subcellular organelles, viruses, and extracellular vesicles. The solution is prepared in a physiological buffer with monovalent and divalent salts to maintain ionic strength and osmotic balance during centrifugation.

Suitable for use in density-based separations requiring gentle conditions compatible with downstream molecular and biochemical analyses, including nucleic acids, proteins, and lipid particles.

  • 1X PBS
  • 1mM Magnesium Chloride
  • 2.5mM Potassium Chloride
  • 1M Sodium Chloride
  • 15% Iodixanol
  • Density Gradient Ultracentrifugation: Forms iso-osmotic gradients for purification of viral vectors (e.g., AAV, lentivirus), extracellular vesicles, and other nanoparticles.

  • Organelle Isolation: Separates nuclei, mitochondria, endosomes, and other subcellular fractions with minimal osmotic stress.

  • Cushion Centrifugation: Acts as an iodixanol cushion to concentrate and clean viral preparations and vesicles while reducing shear damage.

  • Extracellular Vesicle (EV) and Exosome Purification: Enables high-purity isolation from cell culture media and biofluids via step or continuous gradients.

  • Lipoprotein Fractionation: Resolves VLDL, LDL, and HDL based on buoyant density for lipidomics and cardiovascular research.

  • Membrane Microdomain/Raft Isolation: Preserves membrane integrity during detergent-free separation of lipid rafts and membrane proteins.

  • Protein Complex Stabilization: Maintains near-physiological osmolarity and density during centrifugation to protect labile protein assemblies.

  • Virus and Phage Purification: Provides reproducible banding for purification and titering of non-enveloped and enveloped viruses and bacteriophages.

  • Cell and Nuclei Cleanup: Removes debris and dead cells post-lysis or isolation while preserving morphology and function.

  • Nanoparticle and Liposome Separation: Resolves synthetic nanoparticles and liposomes by buoyant density for formulation and delivery studies.
