pDNA Elution Buffer, pH 7.2, is a buffered saline solution formulated for the elution of plasmid DNA from anion-exchange matrices and similar purification workflows. It is suitable for recovering plasmid DNA from preparations derived from E. coli and other common hosts.

The buffering system maintains near-physiological pH during elution, sodium chloride provides ionic strength to disrupt resin–DNA interactions and promote recovery, and EDTA chelates divalent cations to help limit nuclease activity during handling.

  • 50mM Tris/Tris-HCl
  • 10 mM EDTA
  • 0.85 M Sodium Chloride

  • Plasmid Elution (Anion-Exchange): Elutes pDNA from anion-exchange columns/membranes during midiprep/maxiprep workflows.

  • Endotoxin-Reduced Purification: Used in endotoxin-minimizing pDNA purification protocols for transfection-grade DNA.

  • Escherichia coli Plasmid Recovery: Recovers supercoiled plasmids from Escherichia coli lysates following binding/wash steps.

  • Ion-Exchange FPLC: Serves as the high-salt elution buffer for pDNA on ion-exchange columns in chromatography systems.

  • Membrane/Monolith Elution: Elutes pDNA from membrane or monolith adsorbers in high-throughput purification.

  • Transfection Preparation: Prepares pDNA for mammalian cell transfection workflows (typically followed by desalting/buffer exchange).

  • Short-Term Stabilization: Provides pH buffering and nuclease inhibition (via EDTA) for temporary pDNA storage/handling.

  • Precipitation/Ionic Strength Adjustment: Adjusts ionic strength prior to alcohol precipitation or downstream processing.
