pDNA Elution Buffer, pH 7.2 is a buffered salt solution for the elution of plasmid DNA from common purification matrices, including silica and anion-exchange media.

The pH and ionic strength are set to disrupt DNA–matrix interactions and recover plasmid DNA suitable for routine molecular biology workflows such as restriction digestion, ligation, PCR, cloning, and sequencing.

  • 50mM Tris/Tris-HCl
  • 10 mM EDTA
  • 0.85 M Sodium Chloride
  • Anion-Exchange Plasmid Elution: High-salt elution of plasmid DNA from anion-exchange columns/cartridges (50 mM Tris/Tris-HCl, 10 mM EDTA, 0.85 M NaCl).

  • Midiprep/Maxiprep Workflows: Recovery of pDNA from cleared alkaline lysates of E. coli during gravity-flow or FPLC-based purification.

  • Large Construct Recovery: Efficient elution of low-copy or large vectors (e.g., BACs/cosmids) requiring elevated ionic strength.

  • Endotoxin-Reduction Protocols: High-salt elution/wash steps prior to desalting or buffer exchange for low-endotoxin pDNA preps.

  • DNA Stabilization During Elution: EDTA chelation limits nuclease activity; Tris maintains pH 7.2 for pDNA integrity.

  • Downstream Applications (Post-Desalting): Provides eluted pDNA for cloning, restriction digest, PCR, and sequencing after salt/buffer exchange.
