pDNA Equilibration Buffer, pH 7.2, is a buffered solution for conditioning chromatography matrices prior to plasmid DNA purification. It sets the column environment to a defined pH and ionic strength before sample application.

Applications include equilibration of silica or anion-exchange media used to purify plasmid DNA from cleared alkaline lysates of E. coli cultures. The buffer provides consistent conditions for binding and subsequent processing steps across gravity-flow, spin, or vacuum-based formats.

50mM Tris
10 mM EDTA

Plasmid DNA Column Equilibration: Equilibrates anion-exchange or silica columns/membranes before lysate loading for consistent pDNA binding (50 mM Tris, 10 mM EDTA, pH 7.2).
Endotoxin-Reduced pDNA Workflows: Conditions purification media in transfection-grade plasmid prep to support reproducible flow and binding characteristics.
pDNA Stabilization During Processing: EDTA (10 mM) chelates Mg2+/Ca2+ to inhibit nucleases; Tris (50 mM) maintains pH 7.2 for plasmid integrity.
Dialysis/Buffer Exchange of pDNA: Used as a receiving buffer to transfer plasmid into TE-like conditions for downstream handling and storage.
Resuspension and Dilution of pDNA: Provides a nuclease-inhibiting, pH-stable medium for resuspending pellets or adjusting plasmid concentration.
Escherichia coli Plasmid Prep Workflows: Applied during miniprep/midiprep/maxiprep procedures from Escherichia coli cultures to standardize column conditioning.
