pDNA Equilibration Buffer, pH 7.2, is a ready-to-use buffer intended for equilibrating chromatography media prior to plasmid DNA purification. It is used to establish a stable pH environment on anion-exchange or silica-based columns before loading cleared lysates.

Typical applications include preparing gravity-flow, spin, or FPLC columns for processing plasmid DNA from E. coli cultures. The buffer is applied after column preparation or regeneration and before wash and elution steps to maintain consistent conditions throughout the workflow.

  • 50mM Tris
  • 10 mM EDTA
  • Anion-Exchange Column Equilibration: Pre-equilibrates anion-exchange resins for plasmid DNA purification from E. coli lysates.

  • FPLC/Cartridge Conditioning: Conditions anion-exchange cartridges on systems like ÄKTA prior to sample loading.

  • Lysate Buffer Adjustment: Dilutes and adjusts cleared alkaline lysates to optimal pH/ionic strength before chromatography.

  • Pre-Elution Wash Steps: Used in wash sequences to remove RNA, proteins, and endotoxins prior to plasmid elution.

  • Plasmid Stability During Processing: 50 mM Tris, 10 mM EDTA maintain pH and chelate divalent cations to protect pDNA from nucleases.
