pDNA Wash Buffer, pH 7.2 is a Tris/EDTA and high-ionic-strength wash solution used in plasmid DNA purification workflows to remove residual proteins, salts, and other contaminants from silica-based matrices or anion-exchange resins prior to elution. The buffering capacity at pH 7.2 supports plasmid stability, and chelation of divalent cations limits nuclease activity during wash steps.

Applicable to plasmid preparations from E. coli using spin columns or gravity-flow formats, this buffer aids in disrupting nonspecific interactions while maintaining conditions suitable for plasmid retention on the purification matrix until elution.

  • 50mM Tris
  • 10 mM EDTA
  • 0.6 M Sodium Chloride
  • Plasmid DNA Purification: Wash step in alkaline lysis workflows to clean pDNA using a high-salt Tris/EDTA buffer (50 mM Tris, 10 mM EDTA, 0.6 M NaCl, pH 7.2).
