3M Potassium Acetate, pH 5.5 is a buffering salt solution used in alkaline lysis workflows for plasmid isolation from E. coli. At pH 5.5, potassium ions aid neutralization steps that separate detergent–protein–lipid complexes and high–molecular weight DNA from plasmid DNA.

This solution is used for salt-assisted precipitation of nucleic acids with alcohols and for adjusting ionic strength and pH in molecular biology protocols involving bacterial lysates and purified DNA or RNA.

  • 3M Potassium Acetate
  • Plasmid DNA Miniprep Neutralization: 3M Potassium Acetate (pH 5.5) neutralizes alkaline lysates of E. coli, precipitating SDS, proteins, and genomic DNA while keeping plasmids in solution.

  • RNA Purification: Acidic acetate conditions help remove proteins and DNA contaminants and stabilize RNA during extraction.

  • Nucleic Acid Precipitation: Provides potassium ions and acetate for efficient DNA/RNA precipitation with ethanol or isopropanol.

  • Endotoxin and Debris Removal: Facilitates precipitation of lipopolysaccharides and cellular debris from bacterial lysates (e.g., Escherichia coli).

  • Protein Cleanup: Used to precipitate proteins and reduce polysaccharide carryover in clarified lysates.

  • Acetate Buffer Preparation: Serves as a sterile stock to make acetate buffers at pH 5.5 for biochemical workflows.

  • Enzyme Assays at Mildly Acidic pH: Maintains consistent pH 5.5 conditions for enzymes requiring acetate buffering.

  • Sample Clarification Prior to Chromatography: Aids in removing precipitable contaminants to protect downstream columns.
