Potassium acetate, pH 5.5, is an acetate-buffered salt solution intended for pH control and ionic strength adjustment in molecular biology and biochemistry workflows. It provides an acidic buffering environment suitable for procedures requiring acetate at pH 5.5.

Common uses include neutralization of alkaline SDS lysates during plasmid DNA isolation from E. coli and salt-assisted precipitation of nucleic acids with ethanol or isopropanol. It can also serve as an acetate buffer component in enzyme assays and sample preparation protocols that specify pH 5.5 conditions.

Clear, colorless solution. Store tightly closed at ambient temperature. Inspect for clarity prior to use.

  • Plasmid DNA Purification (Alkaline Lysis Neutralization): 3M Potassium Acetate neutralizes alkaline lysates and precipitates SDS–protein–chromosomal DNA complexes during minipreps from Escherichia coli.

  • Nucleic Acid Precipitation: Supplies potassium ions to promote ethanol/isopropanol precipitation and concentration of DNA and RNA.

  • RNA Cleanup: Enhances selective precipitation of RNA and removal of polysaccharides and protein contaminants.

  • Removal of SDS and Proteins: Facilitates precipitation of SDS and protein contaminants from cell lysates during nucleic acid extraction.

  • Genomic DNA Isolation: Assists salt-dependent precipitation and cleanup steps for high-quality DNA recovery.

  • Acetate Buffering (pH 5.5): Serves as an acetate buffer component at pH 5.5 for assays requiring mildly acidic conditions.

  • Ion-Exchange Chromatography: Used as an acetate buffer/salt at acidic pH for equilibration or elution in anion-exchange workflows.

  • Protein Crystallization Screens: Functions as an acetate salt at pH 5.5 in crystallization condition matrices.
