3M Potassium Acetate, pH 5.5 is a ready-to-use aqueous salt solution for pH control and salt-dependent precipitation in molecular biology and biochemistry. The acetate buffer at pH 5.5 provides conditions suitable for neutralization of alkaline cell lysates and selective precipitation of proteins and detergent complexes during plasmid DNA isolation from E. coli.

Applications include RNA precipitation, DNA cleanup, and use as a buffering component in enzymatic reactions requiring acetate at pH 5.5. Compatible with downstream procedures such as electrophoresis, cloning, and qPCR following appropriate purification.

Supplied as a 3 M solution at pH 5.5. Ready to use; dilute only as required by specific protocols. Store at ambient temperature and protect from contamination. For laboratory use.

  • Plasmid Miniprep Neutralization: 3M Potassium Acetate, pH 5.5 is used in alkaline lysis to neutralize lysates and precipitate SDS, proteins, and chromosomal DNA during plasmid prep from Escherichia coli.

  • DNA/RNA Precipitation and Cleanup: Serves as the salt source to facilitate ethanol or isopropanol precipitation of nucleic acids and removal of contaminants.

  • SDS Removal: Precipitates SDS as insoluble potassium dodecyl sulfate, clarifying samples prior to electrophoresis or spectrophotometric analysis.

  • Acetate Buffering for Enzyme Assays: Provides a defined pH 5.5 environment for acid-active enzymes and nucleic acid–related assays requiring acetate buffer.

  • Nucleic Acid Extraction Workflows: Added after phenol–chloroform extraction to adjust ionic strength and pH, improving DNA recovery and purity.
