E. coli Lysis Solution is an alkaline detergent reagent for cell lysis in plasmid DNA preparation workflows from E. coli cultures. It disrupts cell envelopes and promotes alkaline denaturation of chromosomal DNA and proteins, enabling separation of plasmid DNA following neutralization and clarification.

The solution is suitable for use in standard alkaline lysis procedures and is compatible with downstream purification methods, including silica-based and anion-exchange-based plasmid isolation protocols. Intended for laboratory use with E. coli cultures grown in common rich media.

  • 0.2M Sodium Hydroxide
  • 1.0% (w/v) SDS
  • Plasmid DNA Miniprep (Alkaline Lysis): Rapid lysis of E. coli cells for isolation of plasmid DNA using SDS/NaOH alkaline lysis workflows.

  • Midi/Maxi/BAC Plasmid Preparation: Scalable alkaline lysis of E. coli cultures for high-yield plasmid, cosmid, or BAC purification.

  • Colony PCR Template Prep: Generation of crude lysates from E. coli colonies to provide DNA templates for PCR screening.

  • Sanger Sequencing Template Prep: Quick lysis of E. coli colonies to obtain plasmid-rich lysates for direct sequencing workflows.

  • Cloning and Screening Workflows: Lysis step prior to neutralization/clearing for restriction analysis, ligation verification, and construct validation from E. coli cultures.

  • qPCR/ddPCR Screening: Preparation of crude DNA lysates from E. coli for quantitative PCR-based clone verification.

  • High-Throughput Automation: Plate-based alkaline lysis of E. coli in 96-/384-well formats for parallel plasmid screening.
