E. coli Lysis Solution is an alkaline detergent reagent used for the disruption of E. coli cell membranes and denaturation of chromosomal DNA and proteins. It facilitates the release of plasmid DNA from bacterial pellets prepared from liquid or solid culture.

This solution is used as part of standard alkaline lysis workflows in combination with a subsequent neutralization step. It is suitable for processing cultures of plasmid-bearing E. coli strains grown under antibiotic selection prior to downstream clarification and DNA purification.

0.2M Sodium Hydroxide
1.0% (w/v) SDS

Plasmid DNA Purification (Alkaline Lysis) from E. coli: Provides 0.2 M NaOH and 1.0% (w/v) SDS to lyse cells and denature DNA/proteins during standard plasmid prep.
Miniprep/Midiprep/Maxiprep Workflows: Functions as Solution II in conventional alkaline lysis protocols for plasmid extraction from E. coli.
Cleared Lysate Preparation for Cloning/Sequencing: Ensures complete alkaline SDS lysis prior to neutralization, enabling high-quality plasmid for restriction digest, cloning, and sequencing.
Phagemid/M13 ssDNA Isolation: Used for alkaline SDS lysis of infected E. coli before PEG/NaCl precipitation of single-stranded DNA.
BAC/Cosmid Plasmid Preparation: Applied in gentle alkaline lysis of large plasmids from E. coli with modified handling to minimize shearing.
Rapid Lysis for Screening/qPCR Templates: Enables quick alkaline SDS lysis of E. coli cells to release plasmid DNA for colony PCR or qPCR screening.
