E. coli Lysis Solution is an alkaline detergent reagent used to disrupt E. coli cells and release plasmid DNA during alkaline lysis workflows.

The combined action of a strong base and an anionic surfactant disrupts the cell envelope and denatures chromosomal DNA and proteins, providing lysates suitable for downstream plasmid purification.

  • 0.2M Sodium Hydroxide
  • 1.0% (w/v) SDS
  • Plasmid DNA Extraction (Alkaline Lysis): Rapid alkaline SDS lysis of E. coli cultures for miniprep/midiprep plasmid purification.

  • High-Throughput Plasmid Screening: 96-well/automation-friendly lysis of E. coli for parallel plasmid prep workflows.

  • Colony PCR Preparation: Quick lysis of E. coli colonies to generate DNA templates for PCR screening.

  • Crude Lysate for Sanger/NGS: Preparation of plasmid-containing lysates from E. coli for sequencing workflows.

  • Bacterial Strain Verification: Alkaline SDS lysis of E. coli to obtain DNA for confirming inserts or edits by PCR.

  • Transfection-Grade Plasmid Prep (Upstream Step): Initial alkaline lysis of E. coli prior to downstream plasmid cleanup/polishing.

  • Library Cloning Workflows: Lysis of recombinant E. coli for rapid screening and recovery of cloned plasmid constructs.

  • EndA+ Strain Handling: Robust NaOH/SDS lysis of E. coli strains with nucleases to facilitate plasmid recovery.
