pDNA Resuspension Buffer, pH 8.0, is a Tris-based, EDTA-containing buffer formulated for resuspending plasmid DNA pellets after alkaline lysis workflows. It maintains a stable pH during resuspension and chelates divalent cations to minimize nuclease activity.

Intended for rehydration of precipitated or lyophilized plasmid DNA and for preparing samples for downstream procedures such as restriction digestion, molecular cloning, and PCR. Compatible with standard plasmid purification protocols and suitable for short-term storage of plasmid DNA at refrigerated temperatures.

  • 50mM Tris/Tris-HCl
  • 10 mM EDTA
  • Plasmid DNA Resuspension: Rehydrates and solubilizes purified plasmid DNA pellets for downstream use.

  • Plasmid Storage: Provides pH 8.0 Tris and EDTA chelation (50 mM Tris/Tris-HCl, 10 mM EDTA) to protect DNA from degradation during short- and long-term storage.

  • PCR/qPCR Template Preparation: Serves as a stable diluent for plasmid templates to ensure consistent amplification.

  • Sanger/NGS Sequencing: Maintains DNA integrity and compatibility for accurate sequencing reads.

  • Restriction Enzyme Digestion: Suitable for diluting DNA prior to digestion; buffering capacity supports reliable setup when reaction buffer supplies Mg2+.

  • Cloning and Ligation Workflows: Used to prepare plasmid inserts or vectors for ligation, Gibson/HiFi assembly, and downstream transformations into Escherichia coli.

  • Normalization and Aliquoting: Enables consistent concentration adjustments and aliquot preparation for reproducible experiments.

  • DNA Quantification Standards: Provides a defined matrix for spectrophotometric or fluorometric calibration and sample dilution.

  • Transfection Prep: Common resuspension medium for plasmids destined for mammalian cell transfection workflows.

  • Archiving and Shipping: Protects plasmid DNA stability during sample transport and repository storage, including strains carrying plasmids in E. coli.
