pDNA Resuspension Buffer, pH 8, is intended for rehydrating and stabilizing plasmid DNA pellets obtained after alkaline lysis or column-based purification of cultures such as E. coli. The buffered, chelating environment helps maintain DNA integrity during handling and storage.

This buffer is suitable for routine molecular biology workflows involving plasmid DNA, including restriction enzyme digestion, ligation, PCR, cloning, and sequencing.

  • 50mM Tris/Tris-HCl
  • 10 mM EDTA
  • Plasmid Miniprep Resuspension (Buffer P1 equivalent): Used to resuspend E. coli cell pellets prior to alkaline lysis during pDNA isolation (50 mM Tris/Tris-HCl, 10 mM EDTA supports pH control and nuclease inhibition).

  • Midiprep/Maxiprep Cell Pellet Resuspension: Applied to larger-scale E. coli pellets to ensure uniform lysis and high-quality plasmid yields.

  • pDNA Pellet Reconstitution: Dissolves ethanol-precipitated or lyophilized plasmid DNA for handling and storage while protecting from nuclease activity.

  • Short-Term pDNA Storage: Maintains plasmid integrity at pH 8.0; EDTA chelation minimizes metal-dependent nuclease degradation.

  • Standardization of Plasmid Stocks: Provides a consistent buffer matrix for preparing reference plasmid solutions for cloning and analytical workflows.

  • BAC/Cosmid Preparation: Resuspends large plasmid-bearing E. coli pellets prior to gentle lysis to preserve high–molecular-weight DNA.
