This histidine–sucrose formulation buffer combines histidine buffering in the mildly acidic range with sucrose as a non-ionic stabilizing excipient. Histidine holds pH near 5.5 across the range commonly specified for monoclonal antibody, antibody-drug conjugate, and viral vector formulations, while 8% sucrose supports osmotic balance and protects labile biologics through freezing, freeze-drying, and reconstitution.

The solution arrives sterile in a 1000 mL format and is ready to use without further preparation. It suits final formulation and buffer exchange, dilution and storage of proteins and viral vectors, and lyophilization cycle development where a low endotoxin background is required.

  • 20mM Histidine
  • 8% Sucrose

Antibody and ADC Formulation: This buffer serves as a platform formulation matrix for monoclonal antibodies and antibody-drug conjugates. The 20mM histidine holds pH at 5.5 to limit aggregation and deamidation, and the 8% sucrose stabilizes the protein against interfacial and freeze–thaw stress.

Lyophilization and Reconstitution: The 8% sucrose acts as a glass-forming lyoprotectant that preserves protein structure through the freeze-drying cycle. The 20mM histidine maintains pH during freezing, drying, and rehydration.

Viral Vector Formulation and Storage: The solution stabilizes AAV and lentiviral preparations at mildly acidic pH. Histidine provides the pH 5.5 buffering environment, and sucrose protects capsid integrity and osmolarity during frozen storage.

Buffer Exchange and Diafiltration: Purified biologics can be exchanged into this matrix by ultrafiltration, diafiltration, or dialysis. The ready-to-use format gives a consistent target formulation ahead of fill-finish.

Protein and Enzyme Stabilization: Acid-stable proteins and enzymes retain activity in this buffer because histidine controls pH and sucrose provides conformational stabilization during handling and storage.

Lipid Nanoparticle and Liposome Formulation: The sucrose content delivers cryoprotection and tonicity for nanoparticle and liposome suspensions. Histidine holds the formulation at pH 5.5 throughout preparation and storage.

Formulation Development and Stability Studies: Development teams use this solution as a defined baseline for excipient screening, forced-degradation studies, and comparability work across histidine–sucrose systems.

Chromatography Elution and Polish Steps: The buffer suits process steps that call for a mildly acidic, low-ionic-strength environment following affinity or ion-exchange capture.

Low-Endotoxin Workflows: The sterile, low-endotoxin format supports cell-based assays, preclinical material preparation, and downstream bioprocessing where endotoxin burden must stay controlled.

Diluent for Biologic Drug Product: The solution functions as a matched-matrix diluent for potency, purity, and binding assays when the sample is already formulated in histidine and sucrose.
