AAV AEX Column Neutralization Solution is used to adjust pH and ionic strength of anion-exchange chromatography eluates containing adeno-associated virus (AAV) capsids. It is added to acidic or low-pH elution fractions immediately after collection to achieve near-neutral conditions suitable for subsequent processing.

This solution provides buffering capacity in the physiological pH range and supports controlled titration to target setpoints for pH and conductivity. It is suitable for workflows that include buffer exchange, polishing steps, and analytical characterization of AAV preparations.

  • 1M Ammonium Acetate

  • AAV AEX Eluate Neutralization: Rapidly neutralizes high-pH AEX elution (e.g., CAPS pH ~10) with 1M ammonium acetate to preserve capsid integrity and infectivity.

  • Preloaded Fraction Collection: Used in collection tubes to ensure each eluted fraction lands at target pH immediately upon contact.

  • Aggregation Prevention: Minimizes AAV capsid aggregation and VP degradation during elution, pooling, and hold steps.

  • UF/DF Conditioning: Adjusts pH and ionic strength to conditions compatible with TFF and buffer exchange workflows.

  • Enzymatic Treatment Prep: Conditions eluate for downstream Benzonase/DNase steps by bringing pH into an enzyme-friendly range.

  • Analytics Readiness: Stabilizes samples for SEC-HPLC, AUC, ELISA, and SDS-PAGE by normalizing pH post-elution.

  • General Ion-Exchange Neutralization: Applicable for rapid neutralization after basic elutions in other viral vector and protein AEX workflows.
