AAV AEX CIP Solution is an alkaline, high-ionic-strength cleaning-in-place reagent for anion exchange chromatography systems used in adeno-associated virus purification. It is used between purification cycles to remove bound impurities, host cell residues, and aggregated biomolecules from column hardware and resin surfaces.

The solution is compatible with common strong anion-exchange ligands and stainless-steel fluid paths at recommended temperatures and contact times. Typical applications include column sanitization, bioburden control, and removal of process carryover prior to subsequent runs. Prepared with reagent-grade components and sterilizing filtered (0.2 µm). Store tightly closed at ambient temperature, protect from atmospheric CO₂, and handle with alkali-resistant personal protective equipment. Before re-equilibration, flush systems with water-for-injection or equivalent until neutral pH and appropriate conductivity baselines are reached.

  • Anion Exchange Column CIP (AAV workflows): Regenerates and sanitizes AEX resins used for AAV purification, leveraging 1M Sodium Hydroxide and 2M Sodium Chloride to remove host-cell proteins, DNA, and aggregates.

  • Chromatography System Sanitization: Cleans and sanitizes chromatography skids, valves, and tubing to reduce bioburden and endotoxin load, inhibiting contaminants such as Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa.

  • Cross-Contamination Mitigation: Minimizes carryover between AAV lots by deactivating residual viral particles and process impurities along the AEX flow path.

  • Biofilm and Endotoxin Removal: Disrupts biofilms and strips endotoxins from stainless steel and polymeric surfaces through high-pH (1M NaOH) and high-ionic-strength (2M NaCl) cleaning.

  • Nuclease and Adventitious Agent Inactivation: Denatures DNases/RNases and reduces adventitious microbial burden to protect downstream process integrity.

  • Accessory Component Cleaning: Cleans AEX membranes, depth filters, and single-use flow paths post-run to restore performance and extend usable life.

  • pH: Report at 100mM
