AAV Affinity CIP2 Solution, Non-Sterile is a cleaning-in-place reagent for AAV affinity chromatography columns and associated fluid paths. It is intended for routine maintenance between purification cycles to aid in the removal of bound proteins, nucleic acids, and process-derived residues from affinity resins and system surfaces.

This solution is used after product elution and prior to storage or next-cycle equilibration, following the equipment or resin manufacturer’s instructions. It is compatible with common chromatography system materials such as stainless steel and PEEK when used as directed. Thorough post-CIP rinsing with water or buffer until neutral pH and baseline conductivity is recommended before returning the column to operating conditions. Non-sterile; sterilization or sterile filtration may be performed if required by the process.

  • 6M Guanidine Hydrochloride

  • Chromatography Column Cleaning-In-Place (CIP): High-strength 6M guanidine hydrochloride cleaning step for AAV affinity columns to strip tightly bound contaminants.

  • Resin Regeneration/Stripping: Restores AAV affinity resin performance by denaturing and removing fouling proteins, lipids, and carryover from previous runs.

  • Bioburden Reduction and Sanitization: Denatures microbial contaminants, aiding control of Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa in column and system flow paths.

  • Removal of Host Cell Impurities: Efficiently clears host cell proteins, nucleic acids, and endotoxins from AAV affinity media and hardware.

  • System and Flow-Path Cleaning: Cleans pumps, valves, tubing, and manifolds in downstream processing skids using 6M guanidine hydrochloride to reduce fouling.

  • Aggregate and Foulant Clearance: Disrupts and removes protein aggregates and hydrophobic deposits that accumulate during AAV purification.
